Film
In 2004, Kamlesh made his acting debut with the Hindi film Khakee in which he played the role of a police constable, Kamlesh Sawant, starring Amitabh Bachchan and Akshay Kumar.
In 2007, he made his Marathi debut with the film Ek Krantiveer: Vasudev Balwant Phadke.
In 2011, Kamlesh played the role of a police inspector, Kamlesh Sawant, in the Hindi film Force, starring John Abraham.
In 2015, he appeared in the Marathi film Daagdi Chaawl as Kale. In the same year, Kamlesh gained fame in the Hindi film industry for playing the role of a police inspector, Laxmikant Gaitonde, in the film Drishyam, starring Ajay Devgan and Tabu.
Later, he appeared in a few films Shaheed Bhai Kotwal (2020) as Gomaji Patil (Marathi), Sooryavanshi (2021) as Arnab Deshpande and Drishyam 2 (2022) as Laxmikant Gaitonde (Hindi), and Faas (2022) as Baliram (Marathi).
Television
In 2002, Kamlesh played the role of Rastogi in the Hindi TV series CID on Sony TV. In 2011, he appeared in the Marathi TV show Lakshya in which he played the role of a hawaldar, Maruti Jagdale, on Star Pravah.
In 2016, Kamlesh appeared as Ranjit Shinde in the Marathi television show Nakushi – Tarihi Havihavishi on Star Pravah. In 2022, he played the role of Ramesh in the Marathi TV show Vaastu Rahasya.
Award
Kamlesh was awarded the Best Actor Award in the comedy genre for the Marathi film Zhalla Bobhata at the Maharashtra State Film Awards in 2017.

- In an interview, Kamlesh revealed that after the release of the Hindi film Drishyam in 2015, he received offensive phone calls and abusive text messages from random people, for playing the role of a ruthless police inspector, Laxmikant Gaitonde.
