He joined the ‘Fever 104’ radio channel as an intern a week after his third year final exam. In a few working days, he felt the pressure of work and workload, but he continued to work as an intern for a year. He lost almost 11 kg of body weight due to excessive pressure in the organization. He got his first show “Call Center” as a radio jockey, but the show failed to do well and failed.

During training, he met many Bollywood celebrities including Hrithik Roshan, Jackie Shroff, and Ayushmann Khurrana. Her meeting with Hrithik Roshan was a turning point in her life, as she was met by Sudeep Lahiri who came with Hrithik at the radio station where Koli worked. Sudeep asked Prajakta to start his YouTube channel as he realized that he was capable of making comedy videos.

In February 2015, he started his YouTube channel ‘MostlySane.’ His debut YouTube video was the monologue ‘Five Kinds of Singles on Valentine’s Day.’ His video ‘Funny Sayings Delhiites Use’ went viral in June 2015.
He then started uploading three videos per week including ‘RealTalk Tuesday,’ ‘Comedy Video on Thursday’ and ‘SawaalSaturday.’ He has also uploaded several Vlogs on his YouTube channel. In January 2020, she started the ‘Pretty Fit’ series on her channel featuring Indian celebrities like Neha Kakkar, Kareena Kapoor and Sanya Malhotra.
